There are comics that were known to be printed, but no copies have survived (as far as we know), so those would be the rarest.

As far as Batman, Superman, and Spider-Man, you got two of the first appearances correct.

Batman #1 isn't the first Batman.  That happened in Detective Comics #27 in 1939.  CGC has graded 74 copies.

The first appearance of Superman is in Action Comics #1 (1938), CGC has graded 78 copies.

Batman #1 is from 1940, but it is about the tenth appearance of Batman. CGC has graded 280 copies.

The first appearance of Spider-Man is Amazing Fantasy #15 (1962), CGC has graded 3,497 copies.

Detective Comics #27 and Action Comics #1 probably have similar numbers in existence.  I don't think "rarest" is something that can be said for one or the other on those two books.

Can't find the original Flee Thread, but I was more thing along these lines..
