In short, yes.

But there's other factors affecting its value including the overall condition of the book and buyer's demand for it.

A loose staple will lower the grade overall.  The value will be based on the collective grade. 

How much it lowers the grade will depend on the overall condition of the book.

As already stated above, books are graded in their entirety, I would suggest you take clear pics of the books front and back covers, spine, inside front and rear covers/splash pages and the centerfold and post them in the...https://boards.cgccomics.com/forum/42-hey-buddy-can-you-spare-a-grade/ section to get an overall grade, however, just for staples alone...

NM 9.4 - Staples are generally centered; may have slight discoloration.

NM- 9.2 -  Staples may show some discoloration.

VF/NM 9.0 - Staples may show some discoloration. Only the slightest staple tears are allowed.

VF 8.0 - Staples may show some discoloration. Very slight staple tears, Centerfold is mostly secure. 

FN/VF 7.0 - Staples may show some discoloration. Slight staple tears, Centerfold is mostly secure. 

FN 6.0 - Minor staple tears as well as minor rust migration, Centerfold may be loose.

VG/FN 5.0 - Minor staple tears as well as minor rust migration, Centerfold may be loose. 

VG 4.0 - Staples may be discolored. Minor to moderate staple tears as well as some rust migration, Cover may be loose but not completely detached. Centerfold may be loose or detached at one staple.

GD/VG 3.0 - Staples may be rusted or replaced. Minor to moderate staple tears as well as some rust migration, Cover may be loose or even detached at one staple.  Centerfold may be loose or detached at one staple.

GD 2.0 -  Staples may be degraded, replaced or missing. Moderate staple tears replaced or missing as well as rust migration, Shows substantial wear, Cover may even be detached, Centerfold may be loose or detached. 

FR/GD 1.5 - Staples may be degraded, replaced or missing. Staple tears and stress lines are common, as well as rust migration, Cover may be detached, Centerfold may be loose or detached.

I seriously doubt that we have a "loose staple" here.  The cover may be loose-but-attached at a staple due to cover tears at the legs of that staple.  Or, the cover may be completely detached at a staple due to weak cover stock or rough handling.  But "loose staples" are exceptionally rare (and almost always due to missing wraps).  :preach:
